Evaluation of Isoflavones as Allelochemicals with Strong Allelopathic Activities of Kudzu Using Protoplast Co-Culture Method with Digital Image Analysis

The inhibitory allelopathic activity of Pueraria montana (Kudzu), and activities two putative allelochemical isoflavones, puerarin daidzein, were evaluated using the protoplast co-culture method with digital image analysis lettuce as a recipient (DIA-PP method). Cotyledon protoplasts Kudzu isolated Cellulase R10 Driselase 20 in 0.6 M mannitol solution. Optimal hormonal condition density for growth surveyed. Medium or isoflavones was 50 μl liquid MS basal medium containing 1 μM 2,4-dichlorophenoxyacetic acid, 0.1 benzyladenine, 3% sucrose, 0.4 mannitol. Protoplast division strongly inhibited by at low (104/ml). Slightly less inhibition on cell wall formation yellow pigment accumulation stages also observed. Puerarin did not inhibit three but slightly stimulated high concentrations. By contrast, aglycon puerarin, 100 μM. Daidzein might be one cause strong Kudzu. Grade compared that other plants including an invader plant their allelochemicals studied DIA-PP method.
